Immune Tolerance In Kidney Transplantation

aNefroloji BD, Ankara Üniversitesi Tıp Fakültesi, ANKARA While the immunosuppressive drugs developed in the past two decades have improved the short-term survival of organ allografts, the effects of these regimens on long-term outcome has not yet been determined. A significant shortcoming of current anti-rejection therapies is that recipients require life-long treatment on an immunosuppressive regimen and are left at greater risk of serious side effects. Thus, it is of paramount importance that tolerance strategies be developed whereby the destructive alloimmune response is stopped, while maintaining general immune competence. There are a number of examples where tolerance is clinical transplantation has been shown to exist. A small fraction of patients who have terminated their maintenance immunosuppression seem to maintain a functioning graft. However, it is unknown how and when does the tolerance develop and which strategy can induce tolerance? Several approaches have been proposed in rodent and non-human primates with limited success including T cell depletion, co-stimulation blockage and chimerism, however, there has been no excellent model defined yet.Keywords: Immune tolerance, kidney transplantationTurkiye Klinikleri J Nephrol-Special Topics 2008, 1:67-72
